Biotechnology contributes 8-10 marks from two chapters and is one of the most process-oriented topics in NEET Biology. Notes must clearly outline the steps of recombinant DNA technology: isolation of the gene of interest, cutting DNA with restriction enzymes (molecular scissors that recognise palindromic sequences like EcoRI recognising GAATTC), insertion into a cloning vector (pBR322 plasmid with ampicillin and tetracycline resistance genes, or Ti plasmid of Agrobacterium for plant transformation), transfer into a host organism, and selection of transformants. PCR (Polymerase Chain Reaction) invented by Kary Mullis amplifies DNA through repeated cycles of denaturation (94 degrees Celsius), annealing (primer binding at 50-65 degrees), and extension (72 degrees using Taq polymerase from Thermus aquaticus). Gel electrophoresis separates DNA fragments by size, with smaller fragments migrating faster through the agarose matrix. Each technique should be noted with its specific conditions, enzymes, and applications.
Biotechnology applications tested in NEET span medicine, agriculture, and diagnostics. In medicine, genetically engineered insulin (humulin) produced in E. coli replaced animal-source insulin for diabetics. Gene therapy for ADA deficiency involves introducing a functional ADA gene into the patient's lymphocytes using a retroviral vector. Transgenic animals serve as models for disease research, and Rosie the cow was the first transgenic cow producing human alpha-lactalbumin-enriched milk. In agriculture, Bt crops express the cry gene from Bacillus thuringiensis producing insecticidal crystal proteins: Bt cotton uses cry1Ac and cry2Ab genes effective against cotton bollworm, while Bt brinjal uses cry1Ab. Golden Rice is engineered to produce beta-carotene addressing Vitamin A deficiency. RNA interference (RNAi) technology has been used to create nematode-resistant tobacco plants. Molecular diagnostic techniques including ELISA and DNA fingerprinting (using VNTR or variable number tandem repeats) are tested for their principles and applications in forensics and paternity testing.
