DNA (deoxyribonucleic acid) is the hereditary material in most organisms, and understanding its structure is fundamental to scoring well in NEET Molecular Biology. Watson and Crick proposed the double helix model in 1953, building on X-ray crystallography data from Rosalind Franklin and Maurice Wilkins. The double helix consists of two antiparallel polynucleotide chains held together by hydrogen bonds between complementary base pairs: adenine with thymine (two hydrogen bonds) and guanine with cytosine (three hydrogen bonds). This base pairing rule, known as Chargaff's rule, means the amount of adenine equals thymine and guanine equals cytosine in any DNA molecule. NEET questions test structural details including the major and minor grooves, the sugar-phosphate backbone, 3-prime to 5-prime directionality, and the concept of DNA packaging through histones forming nucleosomes, which further coil into chromatin fibers and eventually condense into visible chromosomes during cell division.
DNA replication is a semi-conservative process, as demonstrated by the Meselson and Stahl experiment using heavy nitrogen (N-15) and light nitrogen (N-14) with E. coli. NEET frequently tests both the experiment design and the conclusion that each daughter DNA molecule contains one parental and one newly synthesised strand. The replication machinery involves multiple enzymes: helicase unwinds the double helix, single-strand binding proteins stabilise the unwound strands, primase synthesises RNA primers, DNA polymerase III extends the primer with new nucleotides in the 5-prime to 3-prime direction, DNA polymerase I removes RNA primers and fills gaps, and ligase seals the remaining nicks. The asymmetry of replication, with the leading strand synthesised continuously and the lagging strand synthesised discontinuously as Okazaki fragments, is a high-frequency question topic that tests both factual knowledge and conceptual understanding of enzyme function.
