Reading Campbell is not enough — you have to practise under exam conditions. Here is how to build a proper USABO mock-exam routine, where the practice material comes from, and how to read your mock scores against the real cutoff.

Here is a pattern we see every year: a student who knows a lot of biology walks out of the USABO Open Exam having under-performed. Almost always, the problem was not knowledge — it was exam execution under time pressure, and it is entirely fixable with proper mock practice. This guide is how to do mocks the right way.
The USABO Open Exam is 50 questions in 50 minutes — about a minute per question. That format punishes two things that pure content study never trains: pacing (recognising a question you cannot crack quickly and moving on) and stamina (staying sharp through the back half). You only build those under realistic simulation. A student who has done ten timed Opens walks in calm; a student who has only read walks in and gets ambushed by the clock.

A mock done casually is close to worthless. A mock done properly is the highest-leverage practice there is. The routine:
The single biggest mistake is doing mock after mock without the error review in step 3. That trains speed but not accuracy, and it plateaus fast.
Resist the urge to convert a mock score into "am I qualifying?" You cannot, reliably, because the Semifinal cutoff is a percentile set after grading each year (roughly the top 10% of Open takers), not a fixed number you can predict in advance. What your mock scores can tell you honestly:

You can absolutely run your own mock routine with official past papers and the discipline to review every error. Where a structured test series adds value is in the parts that are hard to do for yourself: proctored, exam-condition simulations, and — most of all — someone experienced reviewing your errors with you and telling you the pattern you keep missing. That external error-review loop is the difference between "I did a lot of mocks" and "my accuracy actually went up."
